Microbial Physiology, Biochemistry & Genetics Our research programs mainly focus on systematics of Frankia strains isolated from root nodules of Hippöphae salicifolia D. Don located at high altitude of Eastern Himalayas, North Sikkim, India and heterocystous cyanobacteria. Both of them comprise the largest global suppliers of biologically fixed nitrogen in the environment through the nitrogenase enzyme complex encoded by Nif HDK genes/operon, which has been highly conserved throughout evolution. Our results proved that the frankial strains have the ability to survive efficiently in the rhizosphere due to the production of hydroxamate and catecholate type of siderophore under iron deficient condition. Also, these siderophores minimize the metal-induced inhibition of growth in Frankia. Furthermore, Frankia strains have been morphologically (TEM), physiologically and biochemically (FAME and protein profiling) characterized under controlled, natural and stress conditions. We are detecting the genetic variability and phylogenetic relationships among the Frankia strains, heterocystous cyanobacteria, and between the heterocystous cyanobacteria and Frankia strains based on amplification and sequence analyses of highly conserved 16S rRNA molecule, nitrogen fixing genes (nif HDK) and variable IGS region(s). We are also focusing on different factors regulating acid/alkaline phosphatase activity in cyanobacteria and Azolla-Anabaena system and their potential application as a suitable biofertilizer.
